Purpose of the metabolomic core

Metabolomic core facility aims to develop and apply high-throughput approaches to explore the metabolome for cardiovascular diseases. The metabolome is defined as the study of all metabolites for a given biological system i.e. biological fluids, tissues and cells. The metabolome describes the final response of an organism submitted to an external perturbation which can alter its regulation (genetic alteration, disease, Toxic exposition…).

Metabolites are compounds with low molecular weight such as organic acids, sugars, aminoacids… implied in metabolic regulation. Metabolomic core is dedicated to metabolome analysis in several biological matrices in order to determine the molecular determinants of cardiovascular diseases. Hence, our core develops High Resolution/Accurate Mass (HR/AM) quantitation of metabolites (aminoacids, biogenic amines, vitamins, nitrogen bases and nucleotides). Untargeted approach will be developed sooner within our core. This work will aim to discover new biomarkers of cardiovascular diseases by LC/MS.

  • Q-Exactive (ThermoFisher Scientific®)

 

This UPLC-LC/MS configuration allows versatile applications in several fields

 

  • LC-MS or LC-MS/MS experiments in a compact device (114 x 83 x 91 cm (H x P x L))
  • Sensitive instruments for quantitation (HESI II source and S-lens technologies)
  • Precursor ion selection and isolation at high resolution for MS/MS experiments
  • Orbitrap detection achieving a resolution of 140.000 (FWHM @m/z 200) with an accuracy below the ppm value
  • Profile and fingerprint screening

Le Q-Exactive est un instrument polyvalent contribuant au développement de nombreuses  applications dans la recherche clinique:
•    Système LC-MS ou LC-MS/MS de paillasse (114 x 83 x 91 cm (H x P x L))
•    Sensible (Source H-ESI II + S-Lens) pour les approches de quantification absolu
•    Sélection d’un ion précurseur à haute résolution pour expériences MS/MS
•    Systéme de détection Orbitrap pouvant atteindre 140000 de résolution avec une précision inférieure à la ppm
•    Instrument dédié au profilage, à la découverte et à la validation de biomarqueurs
